Scientists showed that loss of mitochondrial carrier homologue 2 (MTCH2) increases mitochondrial oxidative phosphorylation, triggering hematopoietic stem cell and progenitor entry into cycle. [Nat Commun] Abstract

Transmembrane TNF-α Preferentially Expressed by Leukemia Stem Cells and Blasts Is a Potent Target for Antibody Therapy
The authors demonstrated a role for transmembrane tumor necrosis factor-α (tmTNF-α) in leukemia. To characterize tmTNF-α expression in acute leukemia, normal hematopoietic cells, and non-hematopoietic tissues, they used a monoclonal antibody, termed C1, which specifically recognizes the tmTNF-α domain. C/EBPα Activates Pre-Existing and De Novo Macrophage Enhancers during Induced Pre-B Cell Transdifferentiation and Myelopoiesis
Scientists showed that during pre-B-cell-to-macrophage transdifferentiation, C/EBPα binds to two types of myeloid enhancers in B cells: pre-existing enhancers that are bound by PU.1, providing a platform for incoming C/EBPα; and de novo enhancers that are targeted by C/EBPα, acting as a pioneer factor for subsequent binding by PU.1. The Fanconi Anemia Pathway Is Required for Efficient Repair of Stress-Induced DNA Damage in Hematopoietic Stem Cells
Previously, the authors demonstrated such stress-associated DNA damage was sufficient to completely deplete hematopoietic stem cells and promote severe aplastic anemia (SAA) in the Fanconi anemia (FA) knockout mouse model, which has compromised replication-associated DNA repair. They extended this previous work to show that FA mice do not spontaneously develop a hematopoietic phenotype consistent with SAA, even at extreme old age. [Cell Cycle] Abstract

Impaired Hematopoiesis and Disrupted Monocyte/Macrophage Homeostasis in Mucopolysaccharidosis Type I Mice
Investigators evaluated alterations in hematopoiesis and its implications on the production of mature progeny in a mucopolysaccharidosis type I murine model. Despite the significant increase in hematopoietic stem cells, a reduction in common myeloid progenitors and granulocyte-macrophage progenitor cells was observed in Idua-/- mice bone marrow. Non-Hematopoietic and Hematopoietic SIRPα Signaling Differently Regulates Murine B Cell Maturation in Bone Marrow and Spleen
Scientists provide evidence that signal regulatory protein alpha (SIRPα), an Ig-superfamily ITIM-receptor expressed by myeloid but not by lymphoid cells, is involved in regulating B cell maturation. [PLoS One] Full Article

Investigators report the clinical and biological data, and final outcome of 48 pediatric patients with primary-resistant AML enrolled in the Associazione Italiana di Ematologia e Oncologia Pediatrica AML 2002/01 clinical trial. [Br J Haematol] Abstract

Allogeneic Hematopoietic Stem Cell Transplantation Could Improve Survival of Cytogenetically Normal Adult Acute Myeloid Leukemia Patients with DNMT3A Mutations
Researchers retrospectively analyzed the prognostic impact of DNMT3A mutations and explored the role of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in 308 cytogenetically normal acute myeloid leukemia patients who received consolidation of intensive chemotherapy or allo-HSCT. Extramedullary Hematopoiesis Is Associated with Lower Cardiac Iron Loading in Chronically Transfused Thalassemia Patients
Investigators evaluated in a large cohort of chronically transfused patients whether the presence of extramedullary hematopoiesis accounts for the typical patterns of cardiac iron distribution and/or cardiac function parameters. [Am J Hematol] Abstract

Anti-Thymocyte Globulin Could Improve the Outcome of Allogeneic Hematopoietic Stem Cell Transplantation in Patients with Highly Aggressive T-Cell Tumors
Researchers used the anti-thymocyte globulin as the part of the conditioning regimen and evaluated the long-term anti-leukemia effect, the safety and complication in the patients with highly aggressive T-cell lymphomas. [Blood Cancer J] Full Article
