TET1 Is a Tumor Suppressor of Hematopoietic Malignancy
Scientists found that deletion of Tet1 promoted the development of B cell lymphoma in mice. TET1 was required for maintenance of the normal abundance and distribution of 5-hydroxymethylcytosine, which prevented hypermethylation of DNA, and for regulation of the B cell lineage and of genes encoding molecules involved in chromosome maintenance and DNA repair. Ash1l Controls Quiescence and Self-Renewal Potential in Hematopoietic Stem Cells
Researchers report an essential role for the trithorax group gene absent, small, or homeotic 1-like (Ash1l) at this developmental transition. Emergence and expansion of Ash1l-deficient fetal/neonatal hematopoietic stem cells (HSCs) were preserved; however, in young adult animals, HSCs were profoundly depleted. The Aryl Hydrocarbon Receptor Nuclear Translocator Is an Essential Regulator of Murine Hematopoietic Stem Cell Viability
Scientists conditionally deleted the hypoxia inducible factor α (HIF-α) dimerization partner, the aryl hydrocarbon receptor nuclear translocator (ARNT) in the hematopoietic system to ablate activity of both HIF-1α and HIF-2α and assessed the functional consequence of ARNT deficiency on fetal liver and adult hematopoiesis. MYB Controls Erythroid versus Megakaryocyte Lineage Fate Decision through the miR-486-3p-Mediated Downregulation of MAF
To further unravel the molecular mechanisms through which v-myb avian myeloblastosis viral oncogene homolog (MYB) affects lineage fate decision, scientists performed the integrative analysis of miRNA and mRNA changes in MYB-silenced human primary CD34+ hematopoietic progenitor cells. Targeting the Spliceosome in Chronic Lymphocytic Leukemia with the Macrolides FD-895 and Pladienolide B
The authors report studies using FD-895 and pladienolide-B in primary leukemia cells derived from chronic lymphocytic leukemia patients and leukemia-lymphoma cell lines. They found that FD-895 and pladienolide-B induce an early pattern of mRNA intron retention – spliceosome inhibition. [Haematologica]

Potential Role of Notch Signaling in CD34+ Chronic Myeloid Leukemia Cells: Cross-Talk between Notch and BCR-ABL
Researchers studied the relationship between BCR-ABL and Notch signaling and assessed the expression patterns of Notch and its downstream target Hes1 in CD34+ stem and progenitor cells from chronic-phase chronic myeloid leukemia patients and bone marrow from normal subjects. Faster T-Cell Development following Gene Therapy Compared to Haplo-Identical Hematopoietic Stem Cell Transplantation in the Treatment of SCID-X1
Researchers retrospectively analyzed and compared clinical outcomes and immune reconstitution in 13 consecutive severe combined immunodeficiency caused by ILR2G mutation (SCID-X1) patients having undergone haploidentical hematopoietic stem cell transplantation and 14 SCID-X1 patients treated with gene therapy over the same period at a single center level. Mobilization of Hematopoietic Progenitor Stem Cells in Allogeneic Setting with Lenograstim by Subcutaneous Injection, in Daily or Twice-Daily Dosing: A Single-Center Prospective Study with Historical Control
