CDK6 as a Key Regulator of Hematopoietic and Leukemic Stem Cell Activation
Scientists describe a novel role for cyclin-dependent kinase 6 (CDK6) in hematopoietic and leukemic stem cells that exceeds its function as cell-cycle regulator. They found that activation of hematopoietic stem cells requires CDK6, which interferes with the transcription of key regulators including Egr1. [Blood] Abstract

The Cytoskeletal Binding Domain of Band 3 Is Required for Multiprotein Complex Formation and Retention during Erythropoiesis
Researchers used in vitro culture of erythroblasts derived from a patient, and separately shRNA mediated depletion of band 3, to investigate the development of a band 3 deficient erythrocyte membrane and to specifically assess the stability and retention of band 3 dependent proteins in the absence of band 3 during terminal erythroid differentiation. The Notch Driven Long Non-Coding RNA Repertoire in T-Cell Acute Lymphoblastic Leukemia
Scientists aimed to establish a comprehensive compendium of the long non-coding RNA transcriptome under control of Notch signaling. For this purpose, they measured the transcriptional response of all protein coding genes and long non-coding RNAs upon pharmacological Notch inhibition in the human T-cell acute lymphoblastic leukemia cell line CUTLL1 using RNA-sequencing. Design of a 2D No-Flow Chamber to Monitor Hematopoietic Stem Cells
Scientists describe a large array of 2D no-flow chambers allowing the monitoring of single hematopoietic stem cells (HSCs) for several days. This allowed them to stain live human HSCs with fluorescent primary antibodies in order to reveal their stage in the hematopoiesis differentiation pathway. [Lab Chip] Abstract

HDAC1 and Klf4 Interplay Critically Regulates Human Myeloid Leukemia Cell Proliferation
Researchers found that histone deacetylase 1 (HDAC1) expression was negatively correlated with that of Kr├╝ppel-like factor 4 (Klf4) and that acute myeloid leukemia patients with lower HDAC1 level had better prognosis. [Cell Death Dis] Full Article

Latexin Sensitizes Leukemogenic Cells to Gamma-Irradiation-Induced Cell-Cycle Arrest and Cell Death through Rps3 Pathway

Potent Antitumor Activity of the Ad5/11 Chimeric Oncolytic Adenovirus Combined with Interleukin-24 for Acute Myeloid Leukemia via Induction of Apoptosis
Researchers evaluated whether chimeric oncolytic adenovirus-mediated transfer of the human interleukin-24 (hIL-24) gene induces enhanced antitumor potency. They show that the Ad5/11 chimeric oncolytic adenovirus carrying hIL-24 produced high levels of hIL-24 in acute myeloid leukemia cancer cells, as compared with the Ad5 oncolytic adenovirus expressing hIL-24. Multipotent Hematopoietic Progenitors Divide Asymmetrically to Create Progenitors of the Lymphomyeloid and Erythromyeloid Lineages
After showing that HC7 binding does not recognizably affect biological features of human hematopoietic stem and progenitor cells (HSPCs), researchers studied asymmetric cell divisions in different HSPC subtypes and determined the developmental potential of arising daughter cells at the single-cell level. Age-Related Mutations Associated with Clonal Hematopoietic Expansion and Malignancies
Researchers analyzed blood-derived sequence data from 2,728 individuals from The Cancer Genome Atlas and discovered 77 blood-specific mutations in cancer-associated genes, the majority being associated with advanced age. 83% of these mutations were from 19 leukemia and/or lymphoma-associated genes, and nine were recurrently mutated.
