TOP STORY
Cell-State-Specific Metabolic Dependency in Hematopoiesis and Leukemogenesis
By deleting enzymes that affect aerobic glycolysis with different potencies, investigators examined how modulating glucose metabolism specifically affects hematopoietic and leukemic cell populations. [Cell]

PUBLICATIONS (Ranked by impact factor of the journal)
Epigenetic Control of Dendritic Cell Development and Fate Determination of Common Myeloid Progenitor by Mysm1
Using a Mysm1 knockout (Mysm1-/-) mouse model, scientists identified the histone H2A deubiquitinase Mysm1 as a critical regulator in dendritic cell (DC) differentiation. Mysm1-/- mice showed a global reduction of DCs in lymphoid organs whereas development of granulocytes and macrophages were not severely affected. [Blood] Abstract

CXCL12+ Stromal Cells as Bone Marrow Niche for CD34+ Hematopoietic Cells and Their Association with Disease Progression in Myelodysplastic Syndromes
Using immunohistochemistry, scientists show that CXCL12+ cells were located in the cellular marrow or perivascular area, and were in contact with CD34+ hematopoietic cells in control and myelodysplastic syndromes/acute myelogenous leukemia with myelodysplasia-related changes bone marrow. [Lab Invest] Abstract

Pre-Transplant Mobilization with Granulocyte Colony Stimulating Factor Improves B Cell Reconstitution by Lentiviral Vector Gene Therapy in SCID-X1 Mice
Researchers hypothesized that mobilization of hematopoietic stem cell by granulocyte colony-stimulating factor should create temporary space in bone marrow niches to improve engraftment and thereby B cell reconstitution. [Hum Gene Ther] Abstract

Characterization of Transcription Factor Networks Involved in Umbilical Cord Blood CD34+ Stem Cells-Derived Erythropoiesis
Investigators utilized umbilical cord blood CD34+ stem cells induced to undergo erythroid differentiation to characterize the transcriptome and transcription factor networks associated with the γ/β-globin switch during fetal erythropoiesis. [PLoS One] Full Article

Vasculogenic and Hematopoietic Cellular Progenitors Are Scattered within the Prenatal Mouse Heart
Scientists characterized phenotypic profiles and location of putative vasculogenic and hematopoietic cellular progenitors in the embryonic mouse heart. [Histochem Cell Biol] Abstract | Full Article

CLINICAL RESEARCH

First-Line Treatment for Severe Aplastic Anemia in Children: Bone Marrow Transplantation from a Matched Family Donor vs. Immunosuppressive Therapy
Researchers retrospectively compared the outcomes of children with acquired severe aplastic anemia who received immunosuppressive therapy within prospective trials conducted by the Japan Childhood Aplastic Anemia Study Group or who underwent bone marrow transplantation from an HLA-matched family donor registered in the Japan Society for Hematopoietic Cell Transplantation Registry. [Haematologica]

Hematological Recovery after Autologous Stem Cell Transplantation Is Associated with Favorable Outcome in Acute Myeloid Leukemia
Investigators analyzed the prognostic significance of hematological recovery for neutrophils and platelets, stratifying at day 20 after autologous stem cell transplantation in 88 consecutive and homogeneously treated acute myeloid leukemia patients in first remission. [Br J Haematol] Abstract

Comparative Analysis of Unrelated Cord Blood Transplantation and HLA-Matched Sibling Hematopoietic Stem Cell Transplantation in Children with High-Risk or Advanced Acute Leukemia
Researchers present a clinical comparison of unrelated cord blood transplantation and human leukocyte antigen (HLA)-matched sibling allogeneic peripheral blood stem cell or bone marrow transplantation in children with high-risk or advanced acute leukemia. [Ann Hematol] Abstract

REVIEWS
Early Lymphoid Lesions: Conceptual, Diagnostic and Clinical Challenges
The authors summarize the diagnostic criteria, recent advances in predicting progression and current recommendations for management of the more recently recognized early or indeterminate clonal lymphoid lesions of B-cell, T-cell and NK-cell derivation. [Haematologica] Full Article

New Generation Dendritic Cell Vaccine for Immunotherapy of Acute Myeloid Leukemia
The authors review their extensive studies on the development of a protocol for the generation of dendritic cells with improved immunogenicity and optimized for the use in cell-based immunotherapy. [Cancer Immunol Immunother] Abstract

Regulation of Hematopoietic Stem Cell Aging by the Small RhoGTPase Cdc42
The authors introduce the canonical set of phenotypes associated with aged HSCs, focus on the novel aging-associated phenotype apolarity caused by elevated activity of the small RhoGTPase in aged HSCs, discuss the role of Cdc42 in hematopoiesis and describe that pharmacological inhibition of Cdc42 activity in aged HSCs results in functionally young and thus rejuvenated HSCs. [Exp Cell Res] Abstract
