TOP STORY
Replication Stress Is a Potent Driver of Functional Decline in Aging Hematopoietic Stem Cells
Scientists show that cycling old hematopoietic stem cells in mice have heightened levels of replication stress associated with cell cycle defects and chromosome gaps or breaks, which are due to decreased expression of mini-chromosome maintenance helicase components and altered dynamics of DNA replication forks. Seamless Gene Correction of β-Thalassemia Mutations in Patient-Specific iPSCs Using CRISPR/Cas9 and piggyBac
Investigators used the latest gene-editing tool, CRISPR/Cas9 technology, combined with the piggyBac transposon to efficiently correct the human hemoglobin beta mutations in patient-derived induced pluripotent stem cells (iPSCs) without leaving any residual footprint. [Genome Res] Abstract | Press Release

D-Cyclins Repress Apoptosis in Hematopoietic Cells by Controlling Death Receptor Fas and Its Ligand FasL
The authors report that D-cyclins perform a rate-limiting antiapoptotic function in vivo. They found that acute shutdown of all three D-cyclins in bone marrow of adult mice resulted in massive apoptosis of all hematopoietic cell types. [Dev Cell] Abstract | Graphical Abstract

Downregulation of Prdm16 mRNA Expression Is a Specific Mechanism to Avoid Leukemogenesis during HOXB4-Mediated HSC Expansion In Vivo
To gain further insight into the molecular events underlying HOXB4-mediated hematopoietic stem cell (HSC) expansion, investigators analyzed gene expression changes at multiple time points in LinSca1+c-kit+ cells from mice transplanted with bone marrow cells transduced with a MSCV-HOXB4-ires-YFP vector. [Blood] Abstract

Dynamics of Leukemia Stem-Like Cells Extinction in Acute Promyelocytic Leukemia
Researchers investigated the dynamics of acute promyelocytic leukemia before and during therapy with regard to disease initiation, progression and therapeutic response. [Cancer Res] Abstract

Type II Interferon Promotes Differentiation of Myeloid-Biased Hematopoietic Stem Cells
Researchers report that type II interferon signaling is required, both at baseline and during an animal model of LCMV infection, to maintain normal myeloid development. [Stem Cells] Abstract

Rictor Is Required for Early B Cell Development in Bone Marrow
Scientists demonstrate that the deletion of Rictor induced an aberrant increase in the FoxO1 and Rag-1 proteins in bone marrow (BM) B cells and that this increase was accompanied by a significant decrease in the abundance of B cells in the peripheral blood and the spleen, suggesting impaired development of early B cells in adult mouse BM. Identification of Heme Oxygenase 1 (HO-1) as a Novel Negative Regulator of Mobilization of Hematopoietic Stem/Progenitor Cells
Researchers demonstrate that HO-1 plays an important and heretofore unrecognized role in retention of hematopoietic stem/progenitor cells (HSPCs) in bone marrow (BM) niches by modulating negatively activation of mobilization promoting complement cascade, maintaining stromal derived factor-1 (SDF-1) level in the BM microenvironment and attenuating chemotactic responsiveness of HSPCs to SDF-1 and sphingosine-1 phosphate gradients in peripheral blood. CLINICAL RESEARCH

Transplantation Outcomes for Severe Combined Immunodeficiency, 2000-2009
The Primary Immune Deficiency Treatment Consortium was formed to analyze the results of hematopoietic-cell transplantation in children with severe combined immunodeficiency (SCID) and other primary immunodeficiencies. Researchers collected data retrospectively from 240 infants with SCID who had received transplants at 25 centers during a ten-year period. [N Engl J Med] Abstract | Press Release

Autologous Hematopoietic Stem Cell Transplantation in Neuromyelitis Optica: A Registry Study of the EBMT Autoimmune Diseases Working Party
The European Group for Blood and Marrow Transplantation (EBMT) Autoimmune Diseases Working Party conducted a retrospective survey to analyze disease outcome following autologous stem cell transplantation. [Mult Scler J] Abstract

REVIEWS
Complexity of Hematopoietic Stem Cell Regulation in the Bone Marrow Microenvironment
This review is organized in three main parts and will focus on cellular components of the HSC niche which are potential targets for hormonal signals, then review critical regulatory signals in the HSC niche and finally highlight the emerging role of hormonal and paracrine signals in the bone marrow. GSK and Genmab Announce Positive Interim Result for Phase III Study of Ofatumumab as Maintenance Therapy for Relapsed CLL
GlaxoSmithKline plc and Genmab A/S announced that an Independent Data Monitoring Committee interim analysis of a Phase III study, PROLONG, reached the predefined significance level for efficacy. The study evaluated ofatumumab maintenance therapy versus no further treatment (observation) in patients with relapsed chronic lymphocytic leukemia (CLL) who responded to treatment at relapse.
