TOP STORY
Aberrant Overexpression of IL-15 Initiates Large Granular Lymphocyte Leukemia through Chromosomal Instability and DNA Hypermethylation
The authors showed that prolonged in vitro exposure of wild-type large granular lymphocyte to interleukin-15 (IL-15) results in Myc-mediated upregulation of aurora kinases, centrosome aberrancies, and aneuploidy. [Cancer Cell] Abstract | Press Release

LABORATORY RESEARCH

Maintenance of Hematopoietic Stem Cells through Regulation of Wnt and mTOR Pathways
Scientists report that suppression of the mTOR pathway, an established nutrient sensor, combined with activation of canonical Wnt-β-catenin signaling, allows for the ex vivo maintenance of human and mouse long-term hematopoietic stem cells under cytokine-free conditions. [Nat Med] Abstract

Wt1-Specific T-Cell Responses in High-Risk Multiple Myeloma Patients Undergoing Allogeneic T-Cell Depleted Hematopoietic Stem Cell Transplantation and Donor Lymphocyte Infusions
Researchers examined the significance of WT1-specific T-cells responses in patients with relapsed multiple myeloma and high-risk cytogenetics who were undergoing allogeneic T-cell-depleted hematopoietic stem cell transplantation followed by donor lymphocyte infusions. [Blood]
Abstract

Myelopoiesis Is Regulated by Osteocytes through Gsα-Dependent Signaling
Osteocytes are thought to initiate adaptive bone remodeling responses via osteoblasts and osteoclasts. The authors report that these cells regulate hematopoiesis, constraining myelopoiesis through a Gsα-mediated mechanism that affects G-CSF production. [Blood] Abstract

Implantable Microenvironments to Attract Hematopoietic Stem/Cancer Cells
Scientists described the manufacture and image-guided monitoring of an engineered microenvironment with user-defined properties that recruits hematopoietic progenitors into the implant. [Proc Natl Acad Sci USA] Abstract

Transcription Factor Oct1 Is a Somatic and Cancer Stem Cell Determinant
The authors showed that Oct1 promotes tumor engraftment frequency and promotes hematopoietic stem cell engraftment potential in competitive and serial transplants. [PLoS Genet] Full Article

STAT5 Is Crucial to Maintain Leukemic Stem Cells in Acute Myeloid Leukemias Induced by MOZ-TIF2
MOZ-TIF2 is a leukemogenic fusion oncoprotein that confers self-renewal capability to hematopoietic progenitor cells and induces acute myeloid leukemia with long latency in bone marrow transplantation assays. Investigators report that FLT3-ITD transforms hematopoietic cells in cooperation with MOZ-TIF2 in vitro and in vivo. [Cancer Res] Abstract

Expression of Integrin A2 Receptor in Human Cord Blood CD34+CD38-CD90+ Stem Cells Engrafting Long-Term in Nod/Scid-Il2rγcNull Mice
The authors report that primitive cord blood CD34+CD38-CD90+ stem cells, with the ability to reconstitute NOD/SCID-IL2Rγcnull mice long term, at 24 weeks after transplantation, can be prospectively isolated at an increased purity by using integrin α2 receptor as an additional stem cell marker. [Stem Cells] Abstract

Inhibition of T Cell Protein Tyrosine Phosphatase (TC-PTP) Enhances IL-18-Dependent Hematopoietic Stem Cell Expansion
Researchers report that inhibition of T cell protein tyrosine phosphatase, an enzyme involved in the regulation of cytokine signaling, through gene knockout results in a nine-fold increase in the number of hematopoietic progenitors in murine bone marrow. [Stem Cells] Abstract

Association between B-Cell Receptor Responsiveness and Disease Progression in B-Cell Chronic Lymphocytic Leukemia: Results from Single Cell Network Profiling Studies
B-cell receptor signaling proteins were functionally characterized by single cell network profiling in samples from patients with B-cell chronic lymphocytic leukemia in an exploratory study after stimulation with anti-IgM. [Haematologica] AbstractFull Article

CLINICAL RESEARCH

Physicians Compliance during Maintenance Therapy in Children with Down Syndrome and Acute Lymphoblastic Leukemia
Researchers reviewed Methotrexate/Mercaptopurine maintenance therapy data for children with Down syndrome (DS) treated according to the NOPHO ALL92 or the NOPHO ALL2000 protocols. The five year event-free survival for the 66 DS patients was inferior to the 2602 non-DS patients. [Leukemia] Abstract

Improved Outcome for Children with Non-High Risk Acute Lymphoblastic Leukemia after Using an ALL IC-BFM 2002-Based Protocol in Shanghai, China
The authors report the outcome of 92 non-high risk children with acute lymphoblastic leukemia (ALL) following a Berlin-Frankfürt-Münster (BFM) Intercontinental ALL-based protocol. [Br J Haematol] Abstract

First Pediatric Sickle Cell Disease Patient Receives NiCord® Stem Cell Transplantation in Gamida Cell Pilot Study at Duke University
Gamida Cell Ltd. announced that the first patient, enrolled in its pilot study of NiCord® as an investigational treatment for sickle cell disease, has been transplanted at Duke University in North Carolina. [Gamida Cell Ltd.] Press Release

Geron Announces Non-Binding Letter of Intent with BioTime Regarding Stem Cell Assets
Geron Corporation announced that the company has entered into a non-binding letter of intent (LOI) with BioTime, Inc. and BioTime’s recently formed subsidiary, BioTime Acquisition Corporation (BAC). The LOI contains broad terms of a potential transaction through which Geron would contribute to BAC its intellectual property and other assets related to Geron’s discontinued human embryonic stem cell programs.
