Sphingosine-1–Phosphate Facilitates Trafficking of Hematopoietic Stem Cells and Their Mobilization by CXCR4 Antagonists in Mice
Scientists show that the Sphingosine-1-Phosphate (S1P) gradient between the bone marrow and the blood, and expression of S1P receptor 1, is essential for optimal hematopoietic stem cell (HSC) mobilization by CXCR4 antagonists including AMD3100, and for the trafficking of HSC during steady state hematopoiesis. [Blood] Abstract

LABORATORY RESEARCH

Inhibition of Proteasome Deubiquitinating Activity as a New Cancer Therapy
Investigators show that treatment with b-AP15 inhibited tumor progression in four different in vivo solid tumor models and inhibited organ infiltration in an acute myeloid leukemia model. [Nat Med] Abstract

Allelic Methylation Levels of the Non-Coding VTRNA2-1 Located on Chromosome 5q31.1 Predict Outcome in AML
Here scientists show that a 106 nucleotide non-coding RNA vault RNA2-1 (vtRNA2-1), previously mis-annotated as miR886, could potentially play a role in the biology and prognosis of acute myeloid leukemia (AML). [Blood] Abstract

Purinergic Signaling Inhibits Human Acute Myeloblastic Leukemia Cell Proliferation, Migration and Engraftment in Immunodeficient Mice
Researchers show that adenosine-triphosphate or uridine-triphosphate have strikingly opposite effects on human acute myeloblastic leukemia cells. [Blood] Abstract

Forced TR2/TR4 Expression in Sickle Cell Disease Mice Confers Enhanced Fetal Hemoglobin Synthesis and Alleviated Disease Phenotypes
Because forced expression of TR2/TR4 in murine adult erythroid cells paradoxically enhanced fetal γ-globin gene expression in transgenic mice, investigators wished to determine if forced TR2/TR4 expression in a sickle cell disease model mouse would result in elevated fetal hemoglobin synthesis and thereby alleviate the disease phenotype. Synergistic Induction of p53 Mediated Apoptosis by Valproic Acid and Nutlin-3 in Acute Myeloid Leukemia
Scientists hypothesized that co-inhibition of MDM2 and histone deacetylases, with nutlin-3 and valproic acid would additively inhibit growth in leukemic cells expressing wild type TP53 and induce p53-mediated apoptosis. [Leukemia] Abstract

MEK Inhibition Enhances ABT-737-Induced Leukemia Cell Apoptosis via Prevention of ERK-Activated MCL-1 Induction and Modulation of MCL-1/BIM Complex
Researchers report that, unexpectedly, ABT-737 induces activation of the extracellular receptor activated kinase and induction of MCL-1 in acute myeloid leukemia cells. [Leukemia] Abstract

CPTH6, a Thiazole-Derivative, Induces Histone Hypoacetylation and Apoptosis in Human Leukemia Cells
The effect of 3-methylcyclopentilidene-[4-(4′-chlorophenyl)thiazol-2-yl]hydrazone (CPTH6) on histone acetylation, cell viability and differentiation, cell cycle distribution and apoptosis in a panel of acute myeloid leukemia and solid tumor cell lines has been evaluated. [Clin Cancer Res] Abstract

Cationic Liposome-Mediated CXCR4 Gene Delivery into Hematopoietic Stem/Progenitor Cells: Implications for Clinical Transplantation and Gene Therapy
In this work, scientists investigated the non-viral CXCR4 gene delivery into hematopoietic stem/progenitor cells using the cationic liposome agent IBAfect. [Stem Cells Dev] Abstract

CLINICAL RESEARCH

Long-Term Outcomes Among Older Patients Following Nonmyeloablative Conditioning and Allogeneic Hematopoietic Cell Transplantation for Advanced Hematologic Malignancies
The objective of this study is to describe outcomes of patients 60 years or older after receiving minimally toxic nonmyeloablative allogeneic hematopoietic cell transplantation. [JAMA] Abstract | Press Release

CD68 and CD163 as Prognostic Factors for Korean Patients with Hodgkin Lymphoma
In the present work, researchers performed immunohistochemical analysis of CD163 as well as CD68, in 144 Korean Hodgkin lymphoma patients treated between November 1990 and December 2009 in a single center. [Eur J Haematol] Abstract
